David Scott Mustaine (d. 13 Eylül 1961, Kaliforniya), Megadeth grubunun kurucusu, vokalisti ve gitaristidir. Panic ve Metallica gruplarında gitaristlik yapmış, Megadeth ve MD.45 isimli grupları kurmuştur. Megadeth grubunda hâlen vokalist ve gitaristlik yapmaktadır.
David Mustaine, 13 Eylül 1961'de Emily David ve John Mustaine'in çocuğu olarak La Mesa, Kaliforniya'da dünyaya geldi. Babasının ailesi köken olarak Fransız, Kanadalı, İrlandalı ve Finlandiyalı iken, annesi ise Alman kökenliydi. Mustaine, Yehova'nın Şahitleri inancına uygun olarak yetiştirildi.
Panic, Mustaine'in ilk grubuydu. Grup, davulda Mike Leftwych, basta Bob Evans, ritm gitarda Tom Quecke, vokalde Pat Voeks ve gitarda Mustaine'den oluşuyordu. Panic'in ikinci konserinden hemen sonra Mike Leftwych ve grubun ses teknisyeni bir araba kazasında hayatını kaybetti ve grup dağıldı.
Dave, grubun dağılmasından sonra 1981'de Metallica'ya geçti. Mustaine, grubun bateristi Lars Ulrich'in bir dergiye verdiği ilan sonrası Ulrich ve James Hetfield ile tanıştı ve prova bile yapmadan gruba dahil oldu.
