De Arrasceata debuted for Defensor Sporting in the Uruguayan Primera División in October 2012, four months after his 18th birthday. He was a member of the team as it won the 2013 Clausura championship, and played in the Championship play-off, a 3–1 loss to Peñarol.

After a difficult start for the Brazilian giants, De Arrascaeta secured his place as one of the teams most important players, showing his amazing dribbling and play-making skills, for which gained the nickname "Arrascaneta", for his incredible ability to nutmeg (also known as "caneta" in Brazil) rivals.
